(1993) first evaluated the technique of burn depth measurement using ldi. they found that the commonest error (in four out of 17 cases) was overestimation of burn depth by the clinician. in only two cases did they underestimate burn depth. by comparison, when they used ldi to measure burn depth this was correct in all cases (4 superficial dermal and 13 deep-dermal or sub-dermal burns).    pape et al. (2001) published an audit of the use of ldi in the assessment of burn depth. they reported that the ldi allowed accurate assessment of burn depth in 97% of burns, compared with 73% for clinical observation alone. these findings were similar to those reported by monstrey et al. (2001, abstract) who reported ldi accuracy of 93.3–96.6% compared to 61–67% for clinical evaluation, when ldi scanning was performed at day 3 and day 5 post-burn, respectively.    jeng et al. (2001) stated that use of the ldi enabled them to make earlier and more ob-jective determination of the need to excise and graft burns. in a separate study, pape and byrne (2001) presented evidence that the use of ldi avoided unnecessary surgery, resulting in a reduction of both costs and workload.    in 2002 holland et al. also reported that the moorldi  was an accurate method for the prediction of burn wound healing in children (holland et al., 2002). this study involved 57 children with a mean age of 1 year and 10 months. the sensitivity and the specificity of the moorldi  were higher than clinical evaluation:    sensitivity ldi – 0.90; clinical evaluation – 0.66.     specificity ldi – 0.96; clinical evaluation – 0.71 (holland et al., 2002)     equipment required    the moorldi2-bi package includes the moorldi2-bi  scan head, scan controller, touch-screen panel pc, all mounted on a mobile clinical stand that can be used in the ward, operating theatre, consulting room or special laser room. full installation, clinical training and certification is included together with a premium service contract.    summary    the moorldi2-bi can help the burn surgeon by indicating the healing potential (hp) of burn wounds. the current clinical practice is to report burn depth. for example, some burn surgeons use the latarjet classification (first-degree, superficial second-degree, deep second-degree and third-degree). we have introduced the term healing potential (hp) when interpreting moorldi2-bi burn scan images because healing is the recordable outcome measure relevant to large areas of burn, whereas assessment of depth (using biopsy) is only available at discreet points.    a multi-centre study was carried out using the moorldi2-bi  to analyse the ranges of blood flow that correlated with healing within 3 specified time-periods; by post-burn day 14, between post-burn day 14 and 21, and greater than 21 days.    the hp categories are:    hp 14: the wound has the potential to heal within 14 days of the burn injury    hp 14-21: the wound has the potential to heal between 14 to 21 days post burn    hp >21: the wound will take longer than 21 days to heal    the actual healing time of the wound will, however, depend upon other factors that can prolong wound healing (e.g. wound infection or use of inotropes). thus, the moorldi2-bi can provide an indication of the potential of the wound to heal but the actual healing time is dependent upon other clinical, biological and environmental factors.    use of the moorldi2-bi has been shown to avoid unnecessary surgery in patients with superficial dermal wounds and promote early surgery for deep dermal wounds.    the moorldi2  laser doppler blood perfusion imagers use a near infra-red laser beam (785nm in the standard moorldi2-ir imager) or a visible red 633nm laser beam (moorldi2-vr  imager). in the moorldi2-2λ dual wavelength system 633 and 830nm lasers are combined to allow simultaneous imaging with the two wavelengths.    the imagers are able to map tissue blood flow over areas from 5cm x 5cm up to 50cm x 50cm with 256 x 256 pixel resolution; - each pixel being an actual measurement. at high scan rates, over 64,000 measurements are made in less than 5 minutes.    the moorldi2  is suitable for a wide range of clinical and research investigations and measurement of blood flow in the microcirculation. the moorldi2  range features a ccd camera which produces a colour image of the scanned area, making the positioning and comparison of images easier. the moorldi2-hr  highest resolution version is available with resolution to 100 microns.

    ELISA是酶聯(lián)接免疫吸附劑測定（ Enzyme-Linked Immunosorbnent Assay ）的簡稱。它是繼免疫熒光和放射免疫技術(shù)之后發(fā)展起來的一種免疫酶技術(shù)。此項(xiàng)技術(shù)自70年代初問世以來，發(fā)展十分迅速，目前已被廣泛用于生物學(xué)和醫(yī)學(xué)科學(xué)的許多領(lǐng)域。 　　

 

（一）　原理

　　ELISA是以免疫學(xué)反應(yīng)為基礎(chǔ)，將抗原、牽9體的特異性反應(yīng)與酶對底物的催化作用相結(jié)合起來的一種敏感性很高的試驗(yàn)技術(shù)。由于抗原、抗體的反應(yīng)在一種固相載體──聚苯乙烯微量滴定板的孔中進(jìn)行，每加入一種試劑孵育后，可通過洗滌除去多余的游離反應(yīng)物，從而試驗(yàn)結(jié)果的特異性與穩(wěn)定性。在實(shí)際應(yīng)用中，通過不同的設(shè)計(jì)，具體的方法步驟可有多種。即:用于檢測抗體的間接法（圖a）、用于檢測抗原的雙抗體夾心法（圖b）以及用于檢測小分子抗原或半抗原的抗原競爭法等等。比較常用的是ELISA雙抗體夾心法及ELISA間接法。 　　

 

   ELISA檢測試劑盒組成:

樣品類型（SAMPLE TYPES）：

   標(biāo)本必須為液體，不含沉淀。包括血清、血漿、尿液、胸腹水、腦脊液、細(xì)胞培養(yǎng)上清、組織勻漿等。1ml的全血可得到0.5ml的血清或血漿。每個標(biāo)本量收集體積＝100ul×檢測種類。取材前須向銷售人員索要說明書。

   標(biāo)本處理:

收集標(biāo)本前必須清楚要檢測的成份是否足夠穩(wěn)定。對收集后當(dāng)天進(jìn)行檢測的標(biāo)本，儲存在4℃?zhèn)溆?，如有特殊原因需要周期收集?biāo)本，將標(biāo)本及時分裝后放在-20℃或-70℃條件下保存。避免反復(fù)凍融。標(biāo)本2-8℃可保存48小時，-20℃可保存1個月。-70度可保存6個月。部分激素類標(biāo)本需添加抑肽酶。

   ◇血清：

室溫血液自然凝固 10-20 分鐘后，離心 20 分鐘左右（ 2000-3000 轉(zhuǎn) / 分）。收集上清。如有沉淀形成，應(yīng)再次離心。

   ◇血漿：

應(yīng)根據(jù)試劑盒的要求選擇 EDTA 、檸檬酸鈉或肝素作為抗凝劑，加入 10 ％（ v/v ）抗凝劑 （ 0.1M 檸檬酸鈉或 1% heparin 或 2.0%EDTA.Na2） 混合10-20 分鐘后，離心 20 分鐘左右（ 2000-3000 轉(zhuǎn) / 分）。仔細(xì)收集上清。如有沉淀形 成，應(yīng)再次離心。

   ◇尿液、胸腹水、腦脊液：

用無菌管收集。離心 20 分鐘左右（ 2000-3000 轉(zhuǎn) / 分）。仔細(xì)收集上清。如有沉淀形成，應(yīng)再次離心。

   ◇細(xì)胞培養(yǎng)上清：

檢測分泌性的成份時,用無菌管收集。離心 20 分鐘左右（ 2000-3000 轉(zhuǎn) / 分）。仔細(xì)收集上清。檢測細(xì)胞內(nèi)的 成份時，用 PBS （ PH7.2-7.4 ）稀釋細(xì)胞懸液，細(xì)胞濃度達(dá)到 100 萬 /ml 左右。通過反復(fù)凍融，以使細(xì)胞破壞并放出細(xì)胞內(nèi)成份。離心 20 分鐘左右（ 2000-3000 轉(zhuǎn) / 分）。仔細(xì)收集上清。保存過程中如有沉淀形成，應(yīng)再次離心。

   ◇組織標(biāo)本：

切割標(biāo)本后，稱取重量。加入一定量的 PBS ，緩沖液中可加入 1 μ g/L 蛋白酶抑制劑或 50U/ml 的 Aprotinin （ 抑肽酶）。用手工或勻漿器將標(biāo)本勻漿充分。離心 20 分鐘左右（ 2000-3000 轉(zhuǎn) / 分）。仔細(xì)收集上清置于 -20 度或 - 70 度保存，如有必要，可以將樣品濃縮干燥。分裝后一份待檢測，其余冷凍備用。

   檢測原理：

采用雙抗體夾心ABC-ELISA法

保存溫度：2-8℃保存6個月.

注意事項(xiàng)：

電阻率檢測儀（精度：±0.01 MΩ·cm）

Millipore和PTB（德國國家計(jì)量研究所）合作研發(fā)了新型電阻率檢測儀，采用獨(dú)特的套筒式設(shè)計(jì)，增大電極表面積且減小電極間距，電極常數(shù)可達(dá)到0.01 cm^-1，溫度靈敏度為0.1℃，尤其適合低電導(dǎo)率范圍內(nèi)（<1μS/cm@25℃）超純水在線檢測。該電阻率測量儀能確保用國際通用標(biāo)準(zhǔn)對電阻率進(jìn)行描述，以溫度補(bǔ)償和非溫度補(bǔ)償兩種模式顯示。

該高精度電阻率儀的設(shè)計(jì)符合USP<645>適應(yīng)性要求

 

TOC檢測儀（檢測范圍：1-999ppb 精度：±1ppb）

即使是電阻率達(dá)到18.2MΩ·cm@25℃的超純水也可能含有較高的TOC值，從而影響您的實(shí)驗(yàn)精度。因此我們推薦您選用Millipore A10 TOC檢測儀，在產(chǎn)水過程和不取水期間，都可以自動對TOC進(jìn)行持續(xù)監(jiān)測。以便您隨時查看水中的有機(jī)物含量，從而避免產(chǎn)生有機(jī)污染風(fēng)險。

檢測步驟：

l         TOC 檢測儀使用0.5mL石英管來獲取超純水。

l         當(dāng)A10紫外燈打開時，激發(fā)有機(jī)物發(fā)生氧化。

l         有機(jī)物的最終氧化產(chǎn)物為二氧化碳，溶于水后導(dǎo)致電導(dǎo)率增加。

l         TOC檢測儀中鈦電池可對這種電導(dǎo)率的改變進(jìn)行持續(xù)監(jiān)測，并利用算法確保將電導(dǎo)率變化換算成TOC值。
